Révélé par Danse avec les loups, où il remporta sa première citation à l'Oscar dans le rôle de Kicking Bird, Graham Greene est né dans la Réserve des Six Nations (Ontario). Membre de la tribu Oneida, il travaille d'abord comme dessinateur technique et technicien du génie civil avant de devenir preneur de son pour divers orchestres de rock. Il dirige ensuite un studio d'enregistrement à Hamilton, dans l'Ontario, et se rend en Angleterre pour participer à des expériences de théâtre collectif. De retour au Canada, Greene fait sa première apparition à l'écran dans Running Brave, puis tourne aux côtés d'Al Pacino dans Révolution de Hugh Hudson. On le verra ensuite dans Where the Spirit Lives et Powwow Highway, et après le triomphe de Danse avec les loups, dans Clearcut de Richard Bugajski, Coeur de tonnerre de Michael Apted, avec Val Kilmer et Sam Shepard, Au bénéfice du doute de Jonathan Heap, Maverick de Richard Donner, avec Mel Gibson, Camilla de Deepa Mehta, avec Jessica Tandy et Bridget Fonda, Une journée en enfer de John McTiernan, avec Bruce Willis et Samuel L. Graham Greene a tourné dans plusieurs téléfilms américains dont The Last of His Tribe, avec Jon Voight, O Rugged Land of Gold, pour The Family Channel, Where the Spirit Lives, Cooperstown, Unnatural Causes, Happily Ever After : Fairy Tales for Every Child, La Rivière des hautes plaines, The Broken Chain, Épreuves d'amour en Alaska, La Bête fauve, et s'est produit en guest star dans des séries hollywoodiennes comme «La Loi de Los Angeles» et «Northern Exposure». Lauréat du Gemini Award pour sa participation à la série «Dudley the Dragon», il a également tourné dans des production canadiennes comme «Grey Sky», «North of 60» et «9B».
Greene a mené durant plusieurs années une carrière théâtrale extrêmement active à Toronto, où il remporta en 1989 le prix Dora Mavor Moore pour sa création dans «Dry Lips Oughta Move to Kapuskasing». Il compte aussi à son répertoire des pièces comme «Diary of a Crazy Boy», «Coming Through Slaughter» et «Jessica».
Le Southwest Museum de Los Angeles, un des musées les plus importants consacrés à la culture indienne, lui a décerné en 1992 le Lifetime Achievement Award.
